Making Electrocompetent E. coli Cells

Stratagene UV Crosslinker (located in the Schmidt lab). Prewarm the machine by running it for 2 minutes. Then set the machine for "250". The units on this number are 100 J / m2. This amount of radiation kills 99.9% of the cells for Escherichia coli strain REL606.

A typical colony on a plate has roughtly 5-50 million cells.
